Drug Efficacy Evaluation

We examine causes of cell proliferation and differentiation by drugs to investigate mechanisms of efficiency and toxicity of pharmaceuticals. Using established cell lines, primary cultured cells, and biological samples, we conduct experiments including observation of morphological changes, measurement of cell surface antigens and biochemical parameters in culture supernatants.
We conduct experiments using plate readers for absorbance, fluorescence, chemiluminescence, and electrochemiluminescence measurement with immunoassay techniques. Analyses using flow cytometers with plate sorting system, laser scanning cytometers, laser capture microdissection systems, real-time PCR systems, and DNA microarray analysis systems are also available. Making the most of these advanced analysis systems and technologies, we analyze macroscopic to microscopic effects of pharmaceuticals on cell functions.

Drug Efficacy Parameters and Development of Measurement Methods

We measure drug efficacy parameters of various drugs. Measurement of widely used parameters is available, as well as parameters specific to each drug, such as active GLP-1, DPP IV activity, GST, troponin I, sTfR, MDA, antibodies to protein preparations (IgG, IgE, etc.), cytokine, and hormone.

  • Bioassay Method: lymphocyte stimulation tests, ELISpot assays, etc.
  • EIA/RIA Method: measurement using commercial kits and development/measurement of systems using purchased and prepared antibodies
  • Biacore Method: development/measurement of systems with Biacore T-100
  • Flow Cytometry: detection/measurement of cell surface markers and intracellular cytokines
  • Electrophoresis: SDS-PAGE, Western blotting, and immunofixation electrophoresis (IFE), etc.

We perform measurement of drug concentrations of target materials such as antibody, interferon, cytokine, and hormone in biological samples (blood, urine, etc.) using the following methods.

  • Bioassay Method: neutralization test, titration assay, etc.
  • EIA/RIA Method: measurement using commercial kits and development/measurement of systems using receptors and antibodies.
  • Biacore Method: development/measurement of systems with Biacore T-100 and other systems, and development/measurement of systems to detect antibody to drugs.

We perform cytokine measurement of various species using various measurement kits. Detection of maximum 27 cytokines in humans and 23 or 9 cytokines in rodents is possible with only 50 μL of samples (serum, plasma, etc.) with Bio-Rad BioPlex System. The cytokine analysis is also available with the multiplex assay system (electrochemiluminescence: ECL) made by Meso Scale Discovery (MSD). Protein Expression Profiling by Two-Dimensional Electrophoresis

We perform protein expression profiling with Ettan DIGE (differential gel electrophoresis) system and Typhoon 9400 system. For analysis of protein abundance changes, DeCyder software made by GE Healthcare is used. For identification of proteins, please contact us for more information.
